Organolithium cyclisations are the subject of considerable current interest, and by allowing organilithium compounds to react intramolecularly their high reactivity may be tamed and directed towards useful synthetic ends. Cyclisations of nitrogen-containing compounds are of particular value as pyrrolidines and piperidines occur widely in nature, particularly in those alkaloids and peptides which have neuriactive properties. We have recently found that certain tertiary aromatic amides will undergo a novel stereoselective cyclisation when treated with strong organolithium bases. We intend to study this cyclisation in more depth for two reasons. Firstly, the reaction is mechanistically remarkable and poses several questions, the answers to which will help us to understand this important class of transformations. Secondly, the products are potentially highly versatile intermediates for use in stereoselective synthesis, and the reaction could form the key step in a concise synthesis of some neurotransmitter analogues allied to the natural product kainic acid. These compounds are of biological interest and are implicated in the processes of memory and learning.
