The absence of a universal detector has limited the type of molecules that can be analysed in capillary electrophoresis and high performance liquid chromatography. In a preliminary study, we have demonstrated the use of molecularly specific Raman detection in microchromatography by accessing a silica microcolumn via an optical fibre probe. The detector was found to have a linear response in the concentration range 75 to 7000 ng of injected material. For wide commercial application, however, the detection limit must approach 1 ng. This proposal will enable us to test ideas that we believe will enable us to achieve this goal. Major improvements can be made through the reduction of the capillary diameter and the development of new sampling optics. The relatively new technique of capillary electrochromatography, with its plug-flow analyte profile, should also improve sensitivity. In addition, the optimum solvent and buffer conditions will be delineated for a range of chemical and biological analytes.
