Summary on Grant Application Form
1. Boron neutron capture therapy (BNCT) of cancer relies on the specific absorption of neutrons by boron atoms in or close to the tumor, subsequent events leading to cell death;2. Pioneering work (Dewar) showed that aromatic substances in which a C=C has been replaced by a B-N unit are in many ways indistinguishable from the original substances;3. We propose to synthesise borazaromatic analogues of natural heterocyclic substances and of macrocycles such as phthalocyanines and porphyrins which are known to localise in tumors, arguing that they would mimic their non-borazaromatic counterparts, but be able to participate in neutron capture;4. The synthesis of simply borazaromatic indoles and purines would be followed by a full chemical and physical characterisation; we would then develop their reaction chemistry for the synthesis of analogues of such natural heterocycles as tryptophan, adenine, and guanine.
