The phomactins are 'a family of diterpenes which exhibit PAF antagonistic activity. It is proposed to develop a .flexible synthetic approach to the phomactins which will provide improved access to several members.of,this`' family. Following on from preliminary studies already carried out, it is proposed,to complete a synthesis of a heavily functionalized methylenecyclohexaneand investigate the cyclisation of this to form the skeleton of the.phomactins. Final modifications will lead to phomactins D and E. Subsequent work should lead to the completion of syntheses of phomactin A and Sch.' 49028. This programme should provide improved access to phomactins for studies of their biological activity: -
