Biochemical studies provide the key to understanding and exploiting microorganisms and also, crucially, the control of infectious deseases. There is a critical requirement to develop tools to allow cell biologists to study complex chemistry within individual cells. The propsed research is the culmination of four years EPSRC funded instrument development from which a high sensitivity chemical microscope (ToFSIMS) has evolved with the analytical capability to probe complex chemistry with high magnification and high sensitivity. In collaboration with microbiologists, a three year project is now proposed which plans to advance the instrumentation specifically for cellular analysis and validate ToFSIMS for the first time in microbial cell chemistry research. Funds are requested to provide the necessary hardware (freeze-fracture facilities and essential upgrade to our post-ionisation laser system), for consumables, to support a microbiologist post-doctoral research associate and postgraduate research student and cover technician costs.
