Plant natural products continue to provide excellent drug leads and templates for the pharmaceutical industry, but little attention has been given towards this valuable source of chemical diversity in the area of anti-bacterial agents.It is possible that we may well experience many of the problems of the pre-antibiotic era due to the proliferation of multidrug resistant (mdr) bacteria. Life threatening infections caused by mdr bacteria, in particular methicillin resistant Straphylococcus aureus (MRSA), are considerably difficult to treat due to the limited number of effective antibiotics available. A further feature of MRSA is the ease in which it acquires multiple resistance, whether via efflux mechanisms or biochemical target modification.There is consequently an urgent need for new chemistry to address this threat and this research proposes to characterise natural products that lower resistance associated with clinically isolated strains of MRSA. We have identified a number of plant extracts that significantly lower the minimum inhibitory concentration of standard antibiotics against MRSA strains that are normally highly resistant. It is proposed to isolate, structurally elucidate and assess the activity of these compounds as drug lead candidates for modulators of resistance in MRSA.
