In this programme we will attempt to complete the first asymmetric total synthesis of the powerful antitumour agent halichomycin by a biogenetically -mo( cyclisation strategy that will close all three of its medium-sized ether/hemimacrolactam rings by a novel, one-pot, reaction cascade applied on the branch precursor, pre-halichomycin (1). It is postulated that the highly stereodefined structure of 1 and its arrangement of functionality will permit the respective reactive termini to react in the desired way. Molecular models of 1 indicate that cyclisation should lead to the correct ring-system 6, if successful.
