This proposed collaborative research programme will develop a toolbox approach to high throughput assay systems based on micro-droplets suspended in immiscible support/carrier liquids. The research will focus on unit operations within the broader functions of: droplet generation, droplet transportation, droplet merging and splitting and associated detection technologies. The detection systems will enable the investigation of the droplet behaviour as well as generating appropriate integrated detection for high throughput assays. The research will be facilitated through a combination of microfabrication techniques to generate channel and array structures within glass and polymer substrates. The behaviour of each unit operation will be modeled by micro CFD techniques and verified through experimental data taken from fabricated devices of identical design to the modeled structures. The understanding of the unit operations gained, will lead to the design and construction of more complex combinations of droplet formation and manipulation to realise a series of demonstrator devices. The demonstrators will exemplify the potential of micro-droplet based assays within high throughput application in the pharmaceutical industry. The collaboration with the industrial partner (AstraZeneca) will also enable comparison between novel droplet high throughput systems and their current HTS assay systems.
