This proposal addresses the fundamental area of the structural characterisation of isolated molecular species and as such has potential impact throughout all disciplines in the physical and life sciences. To undertake this program we propose a number of related programs to be undertaken across the prepared within specific diameter ranges under mild conditions. The exemplar materials chosen will include inorganic and organometallic compounds together with proteins and other biologically important classes of molecule. In parallel we propose to investigate two fundamental problems in structure determination using electron microscopy and diffraction. In the first of these approaches to the direct inversion of the recorded image and diffraction data to the projected potential of the specimen will be tackled and in the second several alternative routes to significantly enhanced resolution beyond 1A will be explored. The final program will aim to refine the structural data obtained and in doing so will explore a wide variety of theoretical and computational avenues. Within both of these we anticipate that several advances in data collection will be required including the development of strategies for data acquisition from radiation sensitive materials. Finally we propose to extend traditional 2D data through the collection and analysis of 3D datasets.
